Objective: To evaluate the expression of proliferating activity and Bcl-2 protein property in eutopic endometrium with endometriosis.
Methods: Endometrial tissues were obtained for endometrial biopsy from 2 groups of women. The study groups were 10 samples, which consisted of eutopic endometrial tissue with endometriosis. The remaining 10 samples were control group, which consisted of eutopic endometrial tissue without endometriosis. Expressions of Ki-67 and Bcl-2 protein were immunohistochemically investigated by polyclonal antibody. The computer program (Image pro (+), USA) was used to analysis the staining intensity of tissue samples objectively. Endometrial cells were cultured for 72 hours and the MTT colorimetric assay was used to detect cell proliferation.
Results: In the group of eutopic endometrium with endometriosis, the activity of cell proliferation was significantly increased. The expression of Ki-67 protein and MTT assay were significantly higher than those in the control group. And expression of Bcl-2 protein was significantly higher than those in the control group (P<0.05).
Conclusion: Eutopic endometrium with endometriois showed increasing Bcl-2 protein expression and increasing proliferation. It was believed to be associated with endometrial pathological abnormalities in endometriosis.
목적: 자궁내막증을 가진 여성의 자궁내막세포의 증식도와 BCL-2 단백질의 발현 정도를 알아보고자 하였다.
연구 방법: 자궁내막증을 가진 10명의 자궁내막조직을 실험군으로 자궁내막증이 없는 10명의 자궁내막조직을 대조군으로 하여 Ki-67 단백질과 Bcl-2 단백질의 발현을 면역조직화학염색법으로 염색한 후 컴퓨터 프로그램 (Image pro (+), USA)을 이용하여 객관적으로 발현 정도를 측정하였다. 자궁내막세포는 72시간 배양한 후 MTT 측정을 이용하여 세포 증식도를 살펴보았다.
결과: 자궁내막증의 자궁내막세포에서 대조군에 비하여 Ki-67 단백질 발현이 증가되었으며 Bcl-2 단백질 발현도 증가하였다. 또한 MTT 측정 결과 자궁내막증이 있는 경우 자궁내막세포의 증식도가 의미 있게 증가되었다.
결론: 자궁내막증 있는 자궁내막세포는 세포자연사를 조절하는 Bcl-2 단백질 발현이 증가하고 세포증식도의 증가를 보였으며 이러한 현상은 자궁내막증의 병태생리와 관련이 있을 것으로 생각된다.
